We have recently found that cationic liposomes can be expanded by addition of an aqueous phase. Addition of aqueous contents to liposomes containing stearylamine (SA) and cholesterol (Chol) and composed of a ceramide (Cer), phosphatidylethanolamine (PE), and sphingomyelin (SM) [dioleoyl-SM (DOPS)] resulted in an increase in the liposome size to that of a "cloud". In addition, such aqueous contents could be gradually incorporated into liposomes without rupture of liposomes. This property was dependent on the liposome compositions and contents. Addition of contents of less than 30% to the liposome caused expansion of the liposome without rupture. Contents of more than 30% to the liposome resulted in the rupture of liposomes. Addition of contents to liposomes containing Chol and PE caused an increase in the length of the phospholipid head groups and resulted in expansion of liposomes. Addition of contents to liposomes containing Chol, PE, and SM resulted in expansion of liposomes; however, aqueous contents did not cause expansion in liposomes composed of PE and SM.
